Each deposit request has its own Nagad number and timer. Do not reuse a number from an earlier deposit, and do not send money after the timer has expired; start a new request instead.

Both work. The app is easier because the success screen shows the transaction ID with a copy button, and you can switch straight back to the Megapari tab. The USSD menu on *167# is useful on a basic phone or when mobile data is off. The transaction ID then arrives only by SMS, so copy it from Messages and paste it into the cashier.
Whichever you use, send the amount exactly as entered in the cashier. A different figure cannot be matched to your request and goes to manual review.
Personal account vs merchant payment
Nagad has two kinds of receiving accounts, and the cashier can use either. An ordinary mobile number with a Send Money instruction is a personal account. A merchant number, or a redirect to the orange Nagad gateway, is a merchant payment, which in the app is under Payment, not Send Money. Mixing them up fails or lands the money in the wrong place. Read the instruction line under the number before you open Nagad.
The gateway route also skips the TrxID step: after the PIN, Nagad confirms the payment directly and the balance updates. It is the better choice if you have had trouble copying transaction IDs.
